STRoe deer: A validated forensic STR profiling system for the European roe deer (Capreolus capreolus)

European roe deer (Capreolus capreolus L.) are the most common game species in Europe, hunted for meat and trophies. Forensic investigations involving poaching may often benefit from an individual identification method to link a suspect specific incident. The current paper presents forensically validated DNA profiling system called “STRoe deer”. This consists of 12 novel unlinked tetra-nucleotide short tandem repeat (STR) loci two sexing markers, with allelic ladder facilitate accurate genotyping. Validation results using 513 samples collected single population Swiss Plateau demonstrated successful amplification all 14 as little 0.05 ng DNA. Species-specificity tests showed that other members Cervidae family exhibited partial profiles non-specific peaks, whereas Bovidae just cross-species products. Three different methods calculate match probabilities randomly sampled genotypes resulted median ranging 1.4 × 10−13 2.5 10−5. These accounted possible structure, occurrence null alleles relatedness. Based on these results, we conclude STRoe is robust genotyping should prove valuable tool support criminal investigations.
